Abstract

Introduction:Polyunsaturated fatty acids (PUFAs) are useful in reducing deleterious effects of ischemia-reperfusion (IR). This study was designed to compare the effects of treatment with flaxseed oil, rich in linoleic acid and alpha-linolenic acid, and fish oil containing long PUFAs, eicosapentaenoic acid and docosahexaenoic acid, on IR injury in the rat kidney. Methods:As an experimental study, 32 male Wistar rats were randomly divided into four groups, each 8 rats: Sham, ischemia-reperfusion (IR), flaxseed oil (FLO), and fish oil (FO). Correspondingly, experimental groups were administered saline and flaxseed or fish oil at doses of 0.4 g/kg, through gavage. After 4 weeks, the rats underwent right nephrectomy and 45-min clamping of the left renal arteries followed by 24 h reperfusion. Renal function was assessed by measuring serum creatinine (Cr) and blood urea nitrogen (BUN) levels.The oxidative stress and serum aspartate aminotranferase (AST) activitywere measured. Each kidney was sectioned into two equal parts; one part was used for pathological evaluations after hematoxylin-eosin staining and the other was applied in assaying malondialdehyde (MDA) level. Results: Serum Cr and BUN levels, AST activity, and tissue MDA content increased in the IR group. Both fish and flaxseed oils decreased tissue MDA levels (P< 0.05 vs. IR), but could not lead to a significant reduction in the levels of the renal markers. Histological damages increased following IR were also markedly reduced by these two oils. Conclusion: Flaxseed oil as well as fish oil may provide protection against IR-induced renal injury and oxidative stress and these effects were not significant between the two supplementations.
